The Effects of Antigens and of Phytohemagglutinin on Rabbit Spleen Cell Suspensions by G. Harris,* M.b., and R. J. Littleton

چکیده
Phytohemagglutinin (PHA) was first shown to be an initiator of mitosis in cultures of normal human leukocytes from peripheral blood (1). Evidence has been presented (2) that the responding cells from human blood had the characteristics of lymphocytes and were derived from a nondividing population. This has been confirmed by other studies (3, 4) which showed that small lymphocytes transformed into a more primitive cell capable of division, and similar results have been described with rabbit peripheral blood cells (5). The present studies were concerned with the effects of PHA on the stimulation of cell proliferation and antibody production by rabbit spleen cells and comparison with the effects of antigens which have been shown to stimulate DNA synthesis in cell suspensions from the spleens of preimmunized rabbits (6). The mechanism of action of PHA is not clear although it has been suggested that it works like an antigen (7) but there is no direct evidence for this except for the fact that the responding cells have the morphological characteristics of cells potentially capable of antibody production. While it has been claimed that human cells, stimulated by PHA, can produce antibodies (8, 9) and that rabbit lymph node explants can produce a specific anarnnestic response to antigens in response to PHA (10) no evidence of immunoglobulin synthesis was found in suspensions of rabbit peripheral blood cells responding to this material (11).

The Effects of Antigens and of Phytohemagglutinin on Rabbit Spleen Cell Suspensions
Phytohemagglutinin (PHA) stimulated the rate of DNA synthesis in rabbit spleen cell suspensions. Unlike antigens, previous immunization to PHA was not necessary and the specific response could not be transferred by macrophages, although lymphocytes primed by incubation in PHA were able to stimulate other spleen cells not directly exposed to PHA. When rabbits were stimulated by in vivo immunizat...
متن کاملImmunogens of Brucella Abortus S19 Identified By Two-Dimensional Gel Electrophoresis and Immunoblotting
Background: Lipopolysaccharides (LPSs) and several antigenic proteins of Brucella have been considered for preparation of diagnostic reagents and subunit vaccines. The objective of this study was to identify and compare immunogens of B. abortus S19 which induce humoral immune responses in human, goat and rabbit. Material and Methods: The bacterial whole cell extract was prepared in extraction b...
متن کاملProliferate Resonse to Purified Bordetella pertussis Toxin on Murine Spleen Lymphocytes
A purification procedure of pertussis toxin (PT) from submerged culture of Bordetella pertussis (B.pertussis) strain 134 using adsorption and affinity chromatography was discussed. The yield of the resulting PT was approximately 37.5mg/l of concentrated culture supernatant. The polypeptide pattern of the purified PT was investigated by SDS-PAGE and showed five bands between 11 to 26 KDa using l...
متن کاملEffects of anti-Ia sera on mitogenic responses. II. Differential expression of the Ia marker on phytohemagglutinin and concanavalin A- reactive T cells
Genes mapping in the I region of the H-2 complex control a system of lymphocyte alloantigens (Ia) which are expressed on a subpopulation of T cells and on most B cells. Specific anti-Ia serum in the presence of rabbit complement removed the splenic T-cell subpopulation responsive to Con-A, but did not affect the response to phytohemagglutinin (PHA) or Leucoagglutinin. Antibodies specific for Ia...

Histopathological effects of different sublethal concentrations (1, 2 and 4 mg/l) of diazinon, an organophosphorus pesticide, on gill, liver, spleen and kidney of grass carp (Ctenopharyngodon idella) were studied after 1, 7, 15, 30 and 45 days post-exposing fishes weighting 850 ± 155 g to the toxicant for 12 hours at 18-22ºC. In addition, tissues of nostril were examined for transmission electr...
